When Fungal Spores in Genshin Impact were first introduced as an item drop in The Chasm: Underground Mines, they didn't have much use until the Sumeru region arrived. As of Version 3.3, Dendro Traveler, Tighnari, Nilou, and Nahida all need the Fungal Spore Powder item group, alongside the weapons Tulaytullah's Remembrance, A Thousand Floating Dreams, Wandering Evenstar, Staff of the Scarlet Sands, End of the Line, and Fruit of Fulfillment. It's important to strategize the best path to farming Fungal Spores so these Genshin Impact characters and weapons can be their best.
When farming Fungal Spore Powder in Genshin Impact, it's important not to have any Electro or Pyro characters on the team (so leave any recently pulled Raiden Shoguns at the door). When a fungi enemy is exposed to Electro or Pyro, they enter an activated or scorched state respectively. Instead of guaranteed item drops in the Fungal Spore Powder group, they are more likely to drop items in the Fungal Nuclei group. Currently, only five weapons need Fungal Nuclei items: Tulaytullah's Remembrance, Hunter's Path, End of the Line, King's Squire, and Wandering Evenstar.

Fungi in Sumeru may actually be more common than hilichurls. So long as a player spends some time wandering the region, they'll find some fungi to farm. Screen Rant has narrowed down the top four locations (in no particular order) to hunt for fungi, due to their high concentrations in the area.

The Jadeplume Terrorshroom (the boss needed to build Tighnari, Genshin Impact's first 5-star Dendro character) sits on an elevated platform above a large cavern. If players jump off the root bridge leading to the boss, they'll land in a smorgasbord of fungi. Players can pick off all the fungi groups within the circular cave before teleporting back to the root bridge. There are a few fungi in the tunnel behind the Teleport Waypoint that are also useful to farm.

The cliffs southeast of Sumeru City leading down to the triangular river bend are full of fungi. After spawning into the Teleport Waypoint for the area, there are three clusters of Fungi on the cliffs themselves. There is a large cluster on the southernmost bend of the river, allowing for a range of natural Hydro-based Genshin Impact reactions. This is a great place to farm Fungal Spore Powder in Genshin Impact.

The Chatrakam Cave underground Teleport Waypoint is not displayed on the map until it is unlocked, but it is a great starting point to farm Fungal Spores. Starting from the Teleport Waypoint, players should jump down the large hole featured in the photo and take the western staircase. This will lead players down a path full of fungi toward the cave exit. If players don't have the Teleport Waypoint unlocked, they can begin on the large island and enter the cave system. This requires enough progress in Aranara quests to remove the roots blocking the entrance.

This Sumeru Teleport Waypoint, north of Genshin Impact's giant killer robot, seems mundane at first glance but is actually positioned between three spawn points for fungi, making it one of the best places to farm Fungal Spore Powder in Genshin Impact. Players can make a triangle between all three points and find the Teleport Waypoint at its center. Fungi love to gather around the waterbed in front of the large monument. There are also two small clusters directly south and directly east of the Teleport Waypoint. It's easy to jump in, grab the materials, and head off to the next location without too much running around.
As previously explained, there are a lot more locations to find fungi. Players can dive into any cave system, and they'll find a few hordes of fungi lurking in the shallow water. They are also common enemies in Sumeru's Daily Commissions. So long as a player's team lacks Electro or Pyro characters, they'll soon be drowning in one of Genshin Impact's most important Sumeru resources.
